The segment of U.S. Route 641 from Eddyville to Marion begins at the intersection with U.S. Route 62 in Eddyville. Traveling north for 8 miles, the highway enters Fredonia where it turns west at an intersection with Kentucky Route 70. At Crayne, the highway turns north once again to enter Marion.
In Marion, U.S. Route 641 terminates at the intersection with U.S. Route 60, thus ending the segment between Eddyville and Marion.
