U.S. Route 61 is a major north–south highway in the central United States. The segment between Vicksburg and Rolling Fork, Mississippi, is a two-lane highway for most of its length.
The highway begins at the interchange with Interstate 20 in Vicksburg and travels north through the eastern part of the city. It then leaves Vicksburg at the intersection with U.S. Route 61 Business and continues north through the community of Redwood.
At the interchange with Mississippi Highway 3, the highway turns northwest towards the community of Valley Fork. In Valley Fork, it turns briefly west before resuming its regular northwest course to Onward. Here, it intersects the southern terminus of Mississippi Highway 1 and turns generally north.
Leaving Onward and passing through Issaquena, the highway turns slightly northeast in the town of Cary. Continuing northeast, it intersects MS-826 before entering the town of Rolling Fork, where the segment ends at the intersection with Mississippi Highway 14.
