The segment of U.S. Route 641 from Benton to Murray begins at the intersection with US-641 Spur just south of Benton. Travelling initially southeast, the highway enters Hardin where it meets KY-402 at a Missouri J intersection. Leaving Hardin, the highway turns south to enter Dexter where it intersects Kentucky Route 1346. Continuing south, U.S. Route 641 passes just west of Almo where it crosses Kentucky 464 as it approaches Murray.
Approaching Murray, the highway crosses KY-1824 and KY-1429 at an unsignaled intersection before entering Murray at the intersection with Kentucky Route 80. Entering Murray, the highway continues south as 12th Street with the segment from Benton to Murray ending at the intersection with Kentucky Route 2075.
