Mississippi Highway 315 from Sardis to Rich begins at the intersection with U.S. Route 61 in Sardis. Driving west as Lee Street, the road soon leaves town and begins its descent into the Mississippi River delta, passing through the community of Sledge, where it intersects Mississippi Highway 3.

Leaving Sledge, the road continues west, passing through the community of Birdie before reaching the small community of Rich. The segment of MS-315 from Sardis to Rich ends at the northern terminus of MS-315 at the intersection with U.S. Routes 49 and 51.
