Mississippi Highway 315 from Rich to Sardis begins at the intersection with U.S. Routes 49 and 51 just southeast of Lula in the community of Rich. Travelling generally east, the route passes through the community of Birdie before reaching the intersection with Mississippi Highway 3 in the town of Sledge.

Crossing Route 3, MS-315 soon climbs out of the Mississippi River delta as it nears Sardis. Entering Sardis as Lee Street, the segment of MS-315 from Rich to Sardis ends at the intersection with U.S. Route 61 on the west side of town.
