The highway heads generally north, passing through the communities of Grace and Hampton, where it intersects with MS-16. Continuing northwest, it traverses rural farmland and small communities like Foote before turning north near Erwin. Further north, MS-1 intersects MS-12 before bending slightly northeast and entering Wayside, where it meets MS-438. It then takes a northwest turn towards Swiftwater, intersecting MS-454, before finally turning north to reach Greenville. This section of MS-1 concludes approximately one block south of Bowman Blvd. in Greenville.
